EFFECTIVE UTILIZATION OF FLUORESCENT INDICATORS IN SEED SECURITY FOR ANTI-COUNTERFEITING OF BROAD BEAN SEEDS

Abstract


The work showed that no negative influence was found in seed and seedling growth when seeds soaked in rhodamine B (0.1mg/ml), lissamine rhodamine B (0.3, 0.1mg/ml) and safranine T (0.3, 0.1mg/ml) solutions, as well as seeds coated with rhodamine B (1:20, 1:30). And there were two uptake pathways of fluorescent indicators into broad bean seed, one was through the seed coat and the other was through emerging seedling tissues after germination occurred. Under green (546 nm) and blue (495 nm) light excitation, rhodamine B-labeled seeds displayed bright red and orange fluorescence respectively which was obviously different from the control. Further, the vascular bundles of stem, seedling roots and aerial parts of seedlings treated with RB all emitted brilliant fluorescence for a longer time. It suggests that fluorescent indicators with appropriate concentration could be used as an effective anti-counterfeiting label to rapidly and easily distinguishing true (treated) seeds from fake (untreated) seeds.
