THE EFFECTS OF EXPLANT, STRENGTH OF NUTRIENT MEDIUM AND PLANT GROWTH REGULATORS ON REGENERATION OF IRANIAN SOYBEAN (GLYCINE MAX CV. SAMAN)

Abstract


Soybean (Glycine max L.) is one of the most important crops and it’s known as a rich source of vegetable protein and oil in the world. Genetic transformation of soybean is necessary for improvement of the variety. Saman is one of the commercial varieties of soybean in Iran and developing an efficient plant regeneration protocol is necessary for their transformation programs. In this study, the effect of some important factors, such as, types of explants (cotyledon, hypocotyl and primary leaf), different combinations of plant growth regulators (KIN, BAP, IAA, NAA and IBA) and different strength of MS medium (full strength MS and half strength MS) on seed germination, regeneration, shoots elongation and rooting of soybean cultivar Saman were studied. The results were obtained from analysis of variance (ANOVA) experimental data. The results showed that the maximum percentages of germination of soybean seeds were 95% that obtained in full strength MS medium with completed by 2.00 mg/l BAP. The maximum number of shoot induction per explant (7.1±0.22) was observed in the full strength MS containing 1.5 mg/l BAP and 0.1 mg/l IAA from cotyledon explants. Also, the maximum rate of shoot elongation (65±0.33%) was observed in the full strength MS medium containing 0.5 mg/l GA3 and 0.2 mg/l BAP. For rooting, the results of this study showed that the presence of auxins were necessary for rooting of elongated shoots of Saman cultivar. Also, the results showed that the maximum average number and length of root were produced in half strength MS medium with 1.5 mg/l IBA. Finally, the rooted plantlets were maintained in the growth chamber for acclimatization. After acclimatization, plantlets were transferred into the greenhouse for flowering and pod maturation.
