PREVALENCE OF WHEAT STRIPE (YELLOW) RUST (SR) IN SINDH, PAKISTAN

Abstract


Stripe (Yellow) Rust (SR) is an important fungal disease of wheat, caused by Puccinia striiformis f.sp tritici (Pst). Cool temperature and high humidity support the growth of this pathogen. However, in last few years signs of this fungus were observed in the warmer regions of Sindh. Present study was designed for surveillance of SR disease in Sindh that may provide useful information regarding status of disease incidence and severity, while molecular detection of SR in latent period would have a significant role in disease management. Survey of wheat growing areas in different locations of Sindh for disease incidence and severity of SRD was done in February and March 2018. In February 2018, leave samples with vague symptoms of SR were collected and assessed for Pst infection by designed multiplex PCR with internal control (IC). During first survey vague symptoms of SR was found on all observed varieties with variable disease incidence and severity. Expected product of PCR amplification from the samples with vague symptoms confirmed the presence of SR infection in field. While increased infection of disease was found in all observed varieties during second survey in March 2018. It could be concluded from the survey results that the SRD has changed its pattern and moving to warmer areas. The designed PCR assay could be used for detection of latent Pst infection in commercial wheat fields and screening of resistant wheat varieties
