Cloning and expression analysis of transcription factor CoWRKY1 from Camellia oleifera

Abstract


WRKY transcription factor is involved in the physiological processes such as growth, metabolism, and environmental responses. In this study, the full-length cDNA sequence of WRKY1gene from Camellia oleifera was obtained by RACE method. The cDNA was termed as CoWRKY1 (GenBank accession No. MF289559). The bioinformatics analysis revealed that the full-length cDNA was 1108 bp with a 972 bp open reading frame. The gene encoded 323 amino acid residues. The CoWRKY1 protein was hydrophilic without the transmembrane region. The CoWRKY1 protein harbored one WRKY domain and one C2H2 zinc finger domain, and hence, it belonged to the second members of the WRKY gene family. CoWRKY1 protein was closely related to Eucalyptus grandis EgWRKY21 and Quercus variabilis QsWRKY21. Real-time fluorescence quantitative PCR analysis showed that the expression of CoWRKY was firstly up regulated and then down regulated from May to October. The expression of CoWRKY gene showed a significant positive correlation with the expression of CoSQS gene.The prokaryotic expression of the 36.2kDa CoWRKY1 protein was detected after 4 h induction by 0.5mM IPTG at 37 ℃.
