Determination of genetic instability of the genome in Nicotiana tabacum L. cells and stabilization with sophora extract

Sevil Mamedli
Abstract

The radiation-induced instability of the Nicotiana tabacum L. genome was experimentally studied. An increase in the number of chromosomal aberrations in plants of generations M1 and M2 was revealed. The level of chromosomal aberrations depended on the dose of radiation and the method of pollination of plants, and the anti-radiation activity of the sophora extract Sophora japonica L. was studied in case of sharp gamma radiation of tobacco and wheat seeds. Preliminary treatment of seeds with these preparations led to stabilization of the content of chlorophyll and carotenoid pigments in wheat germ and to a decrease in the number of chromosome aberrations in the apical meristem of tobacco germs Nicotiana tabacum L., obtained from irradiated seeds. Thus, by testing the root meristem of tobacco in the yield parameter XA, we showed that pre-treatment of seeds with Sophora extract resulted in a significant decrease in the yield of XA in gamma irradiation, which indicates the presence of radioprotective and antimutagenic properties in this preparation.
