Abstract

Guava (Psidium guajava L.) is a prominent fruit crop of Pakistan and rest of the world. The present study was done to explore guava germplasm through effective micro-propagation technique and breeding program. The breeding results revealed a significant increase in physio-chemical attributes of guava like fruit weight (114.05 g), diameter (56.27 mm), fruit length (67.99 mm), seed core weight (22.81 g), and total seed weight (3.030 g). The biochemical attributes such as total soluble solids (10.53 %), titratable acidity (1.777 %), TSS: TA (19.61), ascorbic acid contents (285.33 mg/100ml), total sugar (7.92 %) and reducing sugar (4.38 %) were also increased. The protocol was standardized for clonal propagation, and we developed in vitro techniques to propagate guava in tissue culture. Various concentrations of BAP were used to obtain maximum shoot induction and plant regeneration. The result revealed that 3.0 mgL-1 BAP supplemented with MS medium gave maximum shoot induction percentage in both cv. Gola and Surahi. Seasonal effect and best node position (1-3 N & 3-6 N) result showed that Green House Raised Plants (GHRP) of the summer season with 3-6 node position gave the best response in shoot induction in both cultivars as compared to Field Grown Plants (FGP).
