Analysis of genetic diversity among 14 biotype Chionanthus retusus (Oleaceae) in China

Abstract


Chionanthus retusus Lindl. & Paxt. is rare species in China. Information on genetic diversity can assist in tree breeding and genetic improvement, though there is a lack of such data for this specie. Thus, the morphological characters and genetic diversity of 14 biotype C. retusus were evaluated. The length, width, thickness, leaf area, length/width, length × width and leaf shape index of 30 leaves of each biotype were analyzed using analysis of variance (ANOVA). The results showed extremely significant differences regarding for them. The genetic diversity of the 14 biotypes was further investigated by amplified fragment length polymorphism (AFLP) finger printing. Seven primer combinations were screened, and four combinations successfully produced countable fingerprint bands, 71.43% of which were polymorphic. Jaccard's similarity coefficients varied from 0.49 to 0.88 with an average of 0.57. The Shannon Information index (I) value was 0.9641, indicating a high level of genetic diversity for the species. At a similarity coefficient was 71.9%, the accessions were distinctly divided into 12 groups by clustering analysis. This study would be helpful for biotype identification of C. retusus, and the variable characters observed might contribute to the protection and utilization of germplasm for C. retusus.
