Karyotype analysis for date palm (Phoenix dactylifera L.) compared with tissue cultured derived plants

Abstract


The variety Gulistan of date palm (Phoenix dactylifera L.) has been investigated in present study, which is derived through inflorescence morphologically there was no any change. Temporary slides prepared from root tips by Fuelgen method. Cytological investigation revealed that it have 2n=36 chromosome and 18 pairs appear in metaphase profile which shows that the chromosome number 1, 2, 5, 6, 7, 8, 9 & 10 are metacentric, 3 & 12 submetacentric, 4, 11, 13 & 15 subtelocentric and 14, 16, 17 & 18 telocentric in both kinds of plants (Mother and Tissue Culture). There were no significant differences in the chromosome length and centromeric positions between the micropropagated plantlets and the mother plant
