Study on Relevant Gene Expression in the Early Somatic Embryogenesis of Panax ginseng Hairy Roots

Abstract


Panax ginseng hairy roots were used as explants in the study for the induction and subculture of calluses and embryonic calluses. Semi-quantitative PCR and real-time fluorescent quantitative PCR were adopted to analyze the expression of genes including LBD29, MAPK4, MAPK6, WRKY3, WRKY6 and WRKY27, which were relevant to the plant somatic embryogenesis, in the early somatic embryogenesis of Panax ginseng hairy roots. Results showed that: these 6 genes displayed significant differences in the average expression levels in four different developmental stages of the early somatic embryogenesis for Panax ginseng hairy roots. Therefore, it was speculated that these 6 genes were associated with the early somatic embryogenesis of Panax ginseng hairy roots. Moreover, the study also successfully cloned the CDS zone of Panax ginseng gene WRKY6. The sequencing results showed that: the length of Panax ginseng gene WRKY6 was 1041bp, 99.61% similar to the nucleotide sequence of Panax quinquefolium and 97.63% similar to the amino acid sequence of Panax quinquefolium, which laid solid foundations for establishing an efficient and stable system of somatic embryogenesis for Panax ginseng hairy roots.
