MARKER ASSISTED SCREENING OF WHEAT LINES (TRITICUM AESTIVUM L.) FOR DROUGHT TOLERANCE

Abstract


In the current study a total of 30 wheat lines selected from ICARDA were screened for drought tolerance using SSR markers these lines were characterized morphologically under local climatic condition . The morphological parameters include plant height ,leaf length , leaf width , grains number per spike , spike length, grains weight per spike in gram and 1000 grain weight. The maximum plant height was recorded for SERI1B;/KAUZ./HEVO/3/AMAD/4/ATTILA//’PSN/BOW3/.ATTILA (line29) which was 96 cm. The maximum leaf length was observed for MILAN/*DUCULA//*AL-ZEHRAA-*1 (line3) i-e 33.8 cm where as, the maximum leaf width was recorded for SERI.1B*2/3/KAUZ*2 /BOW//KAUZ//4/KAUZ/FLORKWA-1 (line19) which was 2 cm. The maximum spike length was observed for QAFZAH-7/FLAG-4SEKSAKA-7*//SHUHA-3//PGO/*SERI*82 (line12). Line 14 displayed maximum number of grains per spike i-e 72 . The maximum grain weight per spike and 1000 grains weight were recorded for SERI.1B*2/3/KAUZ*2/BOW//KAUZ/*4//KAUZ/*FLORK WA-1 (line20) i-e 2.91 gm and 47.6 gm respectively. The maximum number of alleles were observed for WMC 596 and WMC 9 i-e 5. The maximum number of bands were amplified by WMC 9 i-e 30 while minimum number of bands were amplified by CFA-2123 i-e 21. Primer WMC 173 and WMC 9 showed highest PIC value i-e 0.65. The minimum PIC value was observed for WMC 147 i-e 0.31. Dendrogram was constructed through software which cluster all the wheat lines into 4 groups. Maximum genetic similarity was found among SERI .1 B*2/3/ KAUZ*2-BOW/KAUZ’4/ATTILA’3*BCN and SERI’1B*2/3-KAUZ*2-BOW/-KAUZ/ 4 ,ORKWA,2. On the basis of morphological characters and SSR markers (line2) KAUZ'S-SERI/4/SERI.1B*2/3/KAUZ*2/BOW//KAUZ* and (line7) P1.861/R DWG//PBW343/3/MUNIA/*ALTAR84//AMSEL can be recommended for future testing and to be utilize in breeding programs.
