IN VITRO ANTICANCER MCF-7, ANTICANCER 3T3 CELL LINE, ANTICANCER HELA CELL LINE, ANTI-INFLAMMATORY, BRINE SHRIMP LETHALITY ASSAY (BSLA) AND GC-MS ANALYSIS OF LEAF BUTANOL FRACTION OF RHEUM RIBES (LBFRR)

Abstract


In this study, GC-MS analysis has shown that leaf butanol fraction of rheum ribes (LBFRR) comprises of 10 compounds which exhibited anticancer (MCF-7) and HeLa cell line activities having IC50 values of 29.04± 1.54 and 15.1± 1.5. MTT Assay (MCF-7), MTT Assay (3T3 Cytotoxicity assay), MTT Assay (HeLa cell line), Oxidative Burst Assay using chemiluminescence Technique, B-Hatching Techniques were the methods used for anticancer MCF-7, anticancer 3T3 cell line, anticancer HeLa cell line ,anti-inflammatory and Brine Shrimp Lethality Assay (BSLA). GC-MS was used for structural elucidation. Leaf methanol extract of rheum ribes (LMERR), leaf n-hexane fraction of rheum ribes (LHFRR), leaf aqueous fraction of rheum ribes (LAFRR) were inactive against anticancer (MCF-7) cell line and HeLa cell line. None of extract and fraction of leaf of rheum ribes have shown anticancer 3T3 cell line activities. Leaf methanol extract of rheum ribes (LMERR), leaf n-Hexane fraction of rheum ribes (LHFRR), leaf aqueous fraction of rheum ribes (LAFRR) and leaf butanol fraction of rheum ribes (LBFRR) showed anti-inflammatory activity on ROS having IC50 value of 16.3± 1.7, 23.2± 1.9, 24.5± 2.5 and 21.6± 0.4. Leaf methanol extract of rheum ribes (LMERR) showed Brine shrimp lethality with LD50 332.58 while leaf n-Hexane fraction of rheum ribes (LHFRR), leaf aqueous fraction of rheum ribes (LAFRR) and leaf butanol fraction of rheum ribes (LBFRR) showed no cytotoxicity. This study revealed that leaf butanol fraction of rheum ribes (LBFRR) exhibited significant anticancer MCF-7 and HeLa cell line activities. In the near future, the constituent of leaf butanol fraction of rheum ribes (LBFRR) can be the alternative drug against MCF-7 cell line and HeLa cell line with least toxicity and side effects.
