EVALUATION OF IN VITRO ANTILEISHMANIAL, ANTICANCER 3T3 CELL LINE, HELA CELL LINE ACTIVITIES AND GC-MS ANALYSIS OF WHOLE PLANT N-HEXANE FRACTION OF RHEUM RIBES (WHFRR)

Abstract


In this research study crude extract and fractions of whole plant of rheum ribes were examined for their antileishmanial, anticancer 3T3 cell line and HeLa cell line activities. Antileishmanial bioassay, MTT Assay (3T3 Cytotoxicity assay and MTT Assay (HeLa Cell Lines) were the methods used for antileishmanial, anticancer 3T3 cell line and HeLa cell line activities. GC-MS was used for structural elucidation. The results revealed that GC-MS analysis of whole plant n-hexane fraction of rheum ribes (WHFRR) comprises of 9 compounds which exhibited antileishmanial activity. The inhibitory concentration which is observed for whole plant n-hexane fraction of rheum ribes (WHFRR) against parasitic and vector born disease, Leishmaniasis is (IC50 74.55 ± 0.4 ug/ml). None of extract and fraction of whole plant of rheum ribes shown anticancer 3T3 and HeLa cell line activities. Whole plant methanol extract of rheum ribes (WMERR), whole plant aqueous fraction of rheum ribes (WAFRR), whole plant ether fraction of rheum ribes (WEFRR), Whole plant chloroform fraction of rheum ribes (WCFRR) and whole plant butanol fraction of rheum ribes (WBFRR) exhibited no antileishmanial activity. This study revealed that whole plant n-hexane fraction of rheum ribes (WHFRR) exhibited significant antileishmanial activity. In the near future, the constituents of whole plant n-hexane fraction of rheum ribes (WHFRR) can be the alternative drug against leishmaniasis with least side effects and toxicity.
