Nepeta cataria (Catnip) and Salvia sclarea (clary sage) shoot regeneration and leaf callus cultures interactions with MWCNTs

Abstract


Abstract. Nepeta cataria L. and Salvia sclarea L., are two species of Labiatae family and endangered species in Yazd province of Iran so that they are grown just adjoining of some springs and streams. In this sense we studied MWCNTs effects on shoot regeneration and callus induction of two species for more propagation and better crop. for this purpose we used MS supplemented with 0.1mg/l kinetin (Kin) and 1mg/l naphthalene acetic acid (NAA) as plant growth regulators (PGRs) for callus regeneration of Clary sage and Catnip and for shoot regeneration of them, MS supplemented with indole-3-acetic acid (IAA) (0.1 mg L-1) and 6-benzyl aminopurine (BAP) (1.0 mg L-1). In seed germination of S.sclarea related to the filter paper germinated seeds with 80µg/ml MWCNTs concentration was the highest seedling length and in N.Cataria was 20µg/ml MWCNTs. In the explant cultures, callus mass of S. sclarea with 80µg/ml MWCNTs was 414 mm3, but in the culture media controls was 182.15 mm3 and there was a significant difference between them and in the callus mass of N.Cataria with 20 µg/ml was 304 mm3 and but in the culture media controls was 117.75 mm3 and there was a significant difference between them. In shoot regeneration we showed that the number of shoots in MS along with 80µg/ml MWCNTs was 4.2 per explants and in MS control was 1.85 per explants that has significant difference between them and also in the shoot regeneration of N.Cataria we showed that the number of shoots in MS along with 20µg/ml MWCNTs was 6.17 per explants and in MS controls was 2.12 per explants that there was significant difference between them.
