CHANGES ON THE ENDOGENOUS PHYTOHORMONE CONTENT OF CORNUS WALTERI SEEDS DURING COLD STRATIFICATION AND EFFECT ON

Abstract


Changes of five endogenous hormones in the seeds of Cornus walteri during cold stratification was studied by means of enzyme linked immunosorbent assay (ELISA), the effect of stratification on seeds germination and mechanism were analyzed. The results showed that the IAA content in seeds decreased significantly at the initial stage, but it increased steadily after 30 days. At 120th days, the content reached its peak (65.751 ng·g-1), and it reached 31.141ng·g-1 at 150th day, and then showed an increasing trend,the change of the IAA/ABA was so. The change of the ABA was similar , and basically it showed a downward trend during the whole treatment period, the content reached its peak after 150 days, that was 235.933 ng·g-1. The content change of both GA1/3 and ZRs was relatively stable, nearly showing a straight line. That of iPAs showing two peaks during the whole treatment period,that were respectively 5.937 ng·g-1 (30th day) and 7.124 ng·g-1 (120th day). Correlation analysis showed that there was no significant correlation between different hormone content in seed treatment period, and there was a significant negative correlation between ABA content and GR (p<0.05), and a very significant negative correlation between ABA content and GP (p<0.01). There was significant negative correlation between GA1/3 content and GR, and GP (p<0.05). So both ABA and GA 1/3 play an important regulation role during the seed germination.
