Molecular characterization and recombination analysis of PVY isolates in the north of Iran

Abstract


Potato virus Y (PVY) is one of the most important plant viruses with a wide host range. Tomato, potato, tobacco, eggplant and pepper; PVY isolates have been characterized on the basis of biological, serological and molecular properties in Golestan province in the north of Iran. All PVY isolates responded positively to monoclonal antibodies specific for PVYC, PVYN or PVYO strains, and many of pepper and eggplant PVY isolates reacted to PVYC and PVYN. Also, we found a diversity of recombination patterns along the CP gene of the PVY strains. Due to the identification of recombination in the potato, tomato and eggplant PVY isolates, response to two monoclonal antibodies in these isolates is likely due to recombination between PVY strains in the same plant. Population genetic analysis of PVY was performed based on the CP gene sequences of nine isolates identified in this study and a large dataset from isolates collected across the world. Phylogenetic analysis showed that PVY isolates were clustered in four phylogroups. Some of Golestan PVY isolates were listed in distinct group and three isolates from pepper and potato were placed in PVYN and PVYO groups respectively.
