ANALYSIS OF DOWNREGULATED GENES IN RICE UNDER DROUGHT USING MICROARRAY SYSTEM AND CLONING OF THE PROMOTER REGION OF OsPEAMT2 GENE

Abstract


Drought stress is one of the major abiotic stresses, which affects crops yield and growth worldwide. Differences in gene expression of rice plant in response to drought stress were identified using 44k Rice Oligo microarray. Microarray data showed downregulation of rpL32 gene. The promoter region of rpL32 gene was first cloned into pBluescript vector and was released by restriction of pBluescript vector with Sma1 and EcoR1enzyme and then purified promoter region of rpL32 was further sub-cloned into expression vector. During expression analysis, rpL32 gene showed low expression under Abscisic acid and drought stress. The rpL32 promoter region analyzed revealed different cis-acting elements like TCA element P-box, Skn-1 motif, Sp1 and DREB1A, using PlantCARE software. These were mostly involved in abiotic stress responses, which showed the significance of the promoter region of rpL32 gene involvement during abiotic stresses
