ANALYSIS OF UPREGULATED GENES UNDER DROUGHT STRESS IN RICE AND CLONING OF PROMOTER REGION OF ISOCITRATE LYASE GENE

Abstract


Microarray is a high put technique, used to study differential expression of target genes. This technique can be used for crops that are affected widely by harsh environmental conditions. Rice is one of the crops consumed by a large population all around the world as staple food. Due to scarcity of water, this water logged crop suffers drought stress which results in the reduction of its production. In order to understand the type of genes regulated under drought stress, microarray technique is used. Out of 44,000 genes collectively of microarray, the number of up-regulated genes at day 8 and day 9 were 3701 genes and 2647 genes. These genes were classified into different classes based on biological processes and molecular function. The CKX2 gene involved in panical development was selected from the drought stress microarray data; thus cloning its promoter region in pBluescript vector and the positive clones were used for sequencing. Similarly, the expression analysis of CKX2 gene was checked in the leaves and roots of rice plantlets under drought and ABA stresses which showed the up-regulation of CKX2 gene under drought stress condition. It is suggested that the CKX2 gene promoter can be used to tailor the gene expression of a desirable gene for enhancement of stress tolerance in plants.
