SCREENING OF BACTERIAL STRAINS FOR HEAVY METALS STRESS TOLERANCE, ANTIMICROBIAL POTENTIAL AND OPTIMUM GROWTH AT WIDE RANGE OF pH AND TEMPERATURE

Abstract


The rapidly growing world population demands sustainable and ecofriendly approaches to produce more food. Natural resources such as Plant growth promoting rhizobacteria (PGPR) show great potential to improve plant and soil nutrient status and hence improve plant productivity. In the current study, PGPR were isolated from lower Dir region and screened for their role in antimicrobial potentials against two phytopathogens (Ralstonia solanacearum and Xanthomonas citri) and heavy metals tolerance. Moreover, the growth of these PGPR was also evaluated at wide range of pH and temperature. Among 59 isolates, 15 isolates inhibited growth of Ralstonia solanacearum while 13 isolates showed resistance against Xanthomonas citri respectively. The maximum zone of inhibition was shown by DJ 15 (33.33mm), (18.16mm) against Ralstonia solanacearum, and Xanthomonas citri. For heavy metal tolerance, the highest growth rate (OD) was recorded against lead (2.05), followed by nickel (1.61) and cadmium (1.38). While maximum growth rate was recorded at pH 6 (0.22), followed by pH 8 (0.23) after 3 hour of incubation. Similarly, after 24 hours of incubation the maximum growth rate was recorded at pH 7 (1.38), followed by pH 6 (1.34). The screened isolates showed maximum growth rate recorded at 50 ºC (0.33), followed by 20 ºC (0.29) after 3 hours of incubation. While after 24 hour of incubation the maximum growth rate was recorded at 30 ºC (1.39), followed by 20 ºC (1.36). Overall results revealed that tested PGPR have potential to showed resistance against Ralstonia solanacearum and Xanthomonas citri, toxic heavy metals and can grow at wide range of pH and temperature.
