MOLECULAR CHARACTERIZATION AND PROMOTER ELEMENT ANALYSIS OF GROWTH REGULATOR BRAGRF5 IN BRASSICA CAMPESTRIS L. SSP. CHINENSIS

Abstract
The transcriptional regulation of eukaryotic gene expression is a critical aspect, with transcription factors playing a key role by binding to cis-acting elements in promoter regions. Growth-regulating factors (GRFs) are plant-specific transcription factors. The ectopic expression of Arabidopsis thaliana GRF5 (AtGRF5) resulted in notable increases in the bud formation and transformation efficiency in some crops such as Beta vulgaris and Brassica napus. However, the regulation of GRFs in Brassica campestris L. ssp. chinensis, a significant vegetable crop in China, remains unexplored. In this study, we identified and characterized the AtGRF5 ortholog in B. campestris, named BraGRF5, discovered from BRAD database. Molecular analysis of BraGRF5 gene and exploration of its protein interaction network of BraGRF5 protein were conducted. Therefore, the promoter region of the BraGRF5 gene was scrutinized to identify cis-acting elements, with types and numbers. It is expected to provide a foundation for regulation of BraGRF5, and a beneficial information for improvement of the B. campestris transformation efficiency and molecular breeding.
