GENETIC ANALYSIS OF FERTILITY RESTORATION SYSTEMS AND CYTOPLASMIC TYPES IN ONION (ALLIUM CEPA) GENOTYPES USING DNA MARKERS

Abstract


Comparing the yield of Open-Pollinated Varieties (OPVs) and hybrids in onion underscores the significance of hybrid breeding as hybrids demonstrate superior traits. The hybrid development in onion is based on cytoplasmic genic male sterility system (CGMS). However, identification and maintenance of A (cytoplasmic genic male sterile) and B (maintainer) lines in biennial crop takes four to eight years through conventional breeding approach. The DNA marker based characterization of cytotype and restoration of fertility (Rf) gene reduces the time span and speed up the hybrid development process. The present study was carried out for morphological characterization and identification of type of cytoplasm and genetic makeup at Rf locus. The experiment was conducted at Vegetable Research Institute, Faisalabad and Agricultural Biotechnology Research Institute, Faisalabad. The twenty one onion genotypes were planted for morphological and molecular characterization. The four DNA markers i.e. Cob marker, OrfA501, orf725MFKR1 and orf725MFKR2 were used for cytotype characterization and three markers i.e. AcSKP1FU, AcSKP1SU and AcPMS1 for Rf locus. The genotypes were largely classified into three groups Phulkara, White-Pearl, Sultan, Yellow-S3, Desi-Red-S4, MirpurKhas-S3, Nasarpuri-S4, VRIO-02, Desi-Red, VRIO-10, VRI-06, Golden-ORB and T.E.G. found suitable for derivation of B line. Similarly, White-Pearl-V, 1122, Sultan-F1, HNO300A and Kessar showed potential for derivation of A line while Hike and Glory genotypes were found suitable for derivation of both A and B line. These findings will help in faster derivation of A and B lines which will ultimately speed up the development of hybrid breeding system in onion
