Ameliorative effect of Black Pepper, Turmeric and Ajwa Date in alloxan-induced diabetic rats: formulation optimization and in vivo biological evaluation

Abstract


Globally, diabetes mellitus entailing chronic hyperglycemia has emerged as one of the most serious metabolic disorders challenging public health and well-being which necessitates testing natural antihyperlipidemic materials. This study aims to investigate anti-diabetic effect of black pepper, turmeric and ajwa date pulp+seed and its mixture as a hypolipidemic, antioxidative and protective effect on liver and kidney by study histological changes in alloxanized diabetic rats. Eleven group consisting of 18 male and 18 females  was classified into normal control (non-diabetic), diabetic (positive) control,positive groups receiving Glibenclamide (10 mg/kg of body weight), extract of black pepper (BP), turmeric (T), ajwa pulp (AP), ajwa seeds (AS) and different combinations of BP+T, BP+AP, BP+AS and BP+T+AP+AS. The dose of black pepper aqueous extract was 50 mg/kg/b.w, while dose of rest of materials was500 mg/kg/b.w.  For 08 weeks, the aqueous extract was orally administered once a day. Blood samples were collected at 0, 4th, and 8th weeks of experiment and subsequently serum were separated. Liver and kidney were separated for histopathological studies. Our investigation for phytochemical screening indicated flavonoids, tannins, saponins, steroids and alkaloids present in these extracts. Results of serum glucose, glycosylated hemoglobin, lipid profile and antioxidant biomarkers level revealed significant improvement by AS, BP+AS and mixture of all extracts.  While histological examination also showed protective effects of these extracts. Based on research findings, it was inferred that all tested materials especially ajwa seeds administered solely and in combination with black pepper extract entailed significant hypoglycemic, antihyperlipidemic and weight stabilizing effects in alloxanized diabetic rats and thus might be developed as a promising herbal medicine for diabetes mellitus.
