COMPARATIVE MOLECULAR SCREENING OF MANGIFERA INDICA L. VARIETIES WITH SSR MARKERS

Abstract
Characterization of undiscovered indigenous and exotic mango varieties is required for conservation and future breeding program purposes. The current study was aimed at the molecular characterization of five mango varieties for finding genetic variability and phylogenetic relations among them. DNA was extracted using CTAB method and nanodrop method was followed to quantify DNA. Polymerase chain reaction was performed for DNA amplification with five SSR markers. Four markers produced polymorphic bands while one marker produce monomorphic bands. Mean polymorphic information content, shannon index found was 0.4496, 0.330, respectively. Genepop package of R version 1.1.7 and MEGA version 6.0 were used to analyze data. Genetic variation found was 89% within individuals and 11% between individuals with AMOVA. UPGMA dendrogram and Principal Coordinate Analysis both divided the five varieties into two groups. Three private alleles (205bp, 250bp and 225bp) were also observed in the study.
