OPTIMIZATION OF EXTRACTION OF POLYSACCHARIDES FROM PAEONIA SUFFRUTICOSA SEED MEAL BY RESPONSE SURFACE METHODOLOGY

Abstract
Response surface methodology (RSM) was adopted for the optimization of the extraction and purification of polysaccharides from peony seed meal, and the content of polysaccharides was determined by anthrone-sulfate method. The effects of different extraction conditions on the yield and content of polysaccharides of seed meal were analyzed by single factor experiment. On account of this, response surface design was used to optimize the extraction process of polysaccharides. The optimal extraction process was as follows: extraction temperature 63oC, extraction time 120min, liquid-solid ratio 20:1. Under these conditions, the theoretical predicted value of polysaccharide extraction rate from peony seed meal was 8.665. While, the actual value is 8.54%. This experiment could be used for further development and utilization of Peony seed meal because of the proximity between the hypothetical and original value.
