FTIR Spectroscopy can be an Alternative and Rapid Method for Analyzing the Molecular Structure Variations of Colorectal Carcinoma Cells against Some Fungi Extracts

Abstract


Fungi have been determined to offer protection against cancer side effects, making fungi with approved anticancer properties and their active compounds of great interest. This study investigates the effects of mushroom extracts from Hericium coralloides(M1), Lactarius deliciosus(M2), Lepista nuda(M3), Pleurotus ostreatus(M4), and Suillus collitinus(M5) together on HCT116. Mesenchymal stem cells (MSCs) were used to evaluate the effect on healthy cells. Cell viability was determined by MTT. Dose/response curves were prepared, IC50 values of the compounds were calculated, and the effect of the extracts was compared using this parameter. FTIR spectrophotometer was used to analyze quantitative changes in cellular components in the appropriate spectral range. Evaluation of IC50 values of all fungi species determined that they decreased the cell viability of HCT116. Conversely, no significant reduction in cell viability was detected on MSCs. Changes in cell membrane lipid, protein, and cell nucleic acid ratio between control and mushroom-treated HCT116 were detected by FTIR. The big deal of chemotherapeutic drugs has herbal origins and still, many resources should be investigated for inhibiting the side effects of cancer therapy. The data obtained with this experiment serves as a reference for studies to be a new source of anticancer drugs in modern pharmacology.
