Synergistic Impacts of Traditional Ethnomedicine Loranthus cordifolius Leaf Extracts and Zinc Oxide Nanoparticles on Immune System of Balb/c Mice

Abstract


Plants as traditional herbal medicines have been used by man since his emergence on the planet. Clinical trials and biomedical research has proved that doxorubicin is red devil due to its side effects and toxicities reporting cardiac myopathy, liver and renal problems. In modern times, engineered nanoparticles, nanoemusions and nanotubes are emerging as potent immunomodulatory agents especially due to their green synthesis approach. Similarly traditional medicinal plants are finding new horizons in the field of ethnopharmacology due to development of new purification and application protocols. Present study was aimed to investigate the immuno-stimulatory effects of green synthesised Zinc oxide nanoparticles (ZnONPs) and methanolic leaf extracts of cultural hemiparasitic plant Loranthus cordifolius against doxorubicin induced toxicity in Balb/c mice. Balb/c mice were divided in seven groups having four mice in each group. Group-I served as control group, group-II received 3mg/Kg body-weight (BW) ZnONPs five times/week for eight weeks, group-III received 6mg/Kg BW Doxorubicin injected intraperitoneally thrice a week for eight weeks , group-IV received 3mg+6mg/kg of ZnONPs+ Doxorubicin , group-V received methanolic leaf extracts of L. cordifolius at dosage 300µL/Mice five times/week for eight weeks, group 6 received 300µL+3mg/Kg leaf extracts+ ZnONPs and group 7 received 300µL+3mg/Kg+6mg/kg plant extract+ ZnONPs+ Doxorubicin. The application of ZnONPs stimulated white blood cell count by 103%, body weight of mice by 17%, viability of macrophages by 32 % and percentage of phagocytosis by 58%. However the combined application of leaf extracts and ZnONPs proved more immune-stimulatory as it increased the mentioned parameters by 137%, 27%, 37% and 81%, respectively compared to doxorubicin induced group. Present study recommends the use of ZnONPs and leaf extracts of L. cordifolius in mitigating the toxicity induced by doxorubicin as a result of chemotherapy.
