Effects of individual and combined stresses of Salinity (NaCl) and Heavy metal (NiCl2) Levels on Seed Germination and seedlings of Citrullus colocynthis L.

Abstract


ABSTRACT Salinity and heavy metal stresses are great hindrance for germination and early development of plants. Nickel (Ni) is an indispensible micronutrient for plants at low and toxic in high concentration. However the proficiency depends upon the plants types. Citrullus colocynth is medicinal salt and heavy metal tolerant cucurbit weed. The contemporary target of study is to appraise the influence of single and combined stresses by varying concentrations of sodium chloride and nickel chloride on germination and seedlings of C. colocynth. Germination percentage, radicle length, plumule length, fresh weight and dry weight of seedlings of C. colocynth with three levels of salt (100mM, 200mM and 400mMNaCl), three levels of heavy metal (50, 100, 200uMNiCl2) and Nine combined levels(100mMNaCl +50uM NiCl2), (200mMNaCl +50 uMNiCl2) (400mMNaCl +50uM NiCl2) (100mMNaCl +100uMNiCl2) (200mM NaCl +100uM NiCl2) (400mMNaCl +100uMNiCl2) (100mMNaCl +200uMNiCl2) (200mMNaCl +200 uMNiCl2) (400mMNaCl +200uMNiCl2) were studied. This experiment was conceded out in completely randomized design with four replications. Statistical analysis was performed using 8.1 of STATISTIX software using ANOVA and LSD. The responses to different levels of salinity, heavy metal and combined stresses in all measured traits showed adverse trend with increasing stress levels. This species has suitable germination at 100mMNaCl, 50uM NiCl2 and combined (100mMNaCl +50 uMNiCl2), endure at 200mMNaCl, 100 uMNiCl2, (200mMNaCl +50uMNiCl2), (100mM NaCl +100 uMNiCl2) and (200mM NaCl +100 uM NiCl2). Germination and seedling growth factors declined under high stress levels of salt (400mM NaCl), high heavy metal (100 uM NiCl2) and combined high salt or high heavy metal with any other levels of salt or heavy metal.
