MORPHOLOGICAL AND MOLECULAR DIVERSITY IN OTOSTEGIA FROM IRAN

Abstract


In this study, morphological and molecular diversity of Otostegia populations in the arid regions of Iran were studied. There are three species of Otostegia (O. persica, O. aucheri and O. michauxii) in this region. In this study, thirty characters were studied in eleven populations of these species. In PCA analysis, characters such as plant height, fleshy and juicy leaves and leaf margin were the most important diagnostic characters in intera-generic variation. On the other hand, the characters such as leaf size, leaf apex, number of thorns in the leaf nodes, size of thorns, internode length, fruit length and corolla length are important diagnostic characters in intera-species variation in populations of O. persica. In UPGMA tree of morphological study and NJ tree of molecular analysis, two major clusters were formed. The species were well separated in these clusters. But under the subclusters, populations of these species showed some similarity, except populations of Sarbaz area. By examining the parameters of genetic diversity, the populations of Sarbaz area had the most value of percentage polymorphism, Shanon index and Nei’s genetic diversity. AMOVA analyses indicated significant molecular difference between these species. In the Mantel test, there was no relationship between genetic and geographical distance.
