Introgression of Yr genes for durable yellow rust resistance in high yielding Indian wheat varieties WH711 and PBW343 through marker assisted backcross breeding

Abstract


The present study was undertaken for developing high yielding yellow rust resistant wheat genotypes using marker-assisted selection (MAS). High temperature adult plant resistance gene (Yr36), adult plant resistance gene (Yr18) and all stage plant resistance gene (Yr17) was introgressed from hard red spring wheat line Lassik into high yielding yellow rust susceptible wheat varieties i.e. WH711 and PBW343 using MAS. Foreground selection was carried out using specific markers (WKS1/ASA, csLv34, URIC-LN2) for each genes in BC2F7 and BC2F8 generation. In each backcross generation, background selection was performed using 100 and 101 simple sequence repeat loci for the cross WH711 x Lassik and PBW343 x Lassik. Nine BC2F8 progenies of the cross WH711 x Lassik and five of the cross PBW343 x Lassik having desirable Yr genes, and showing up to 96.48% and 97.50% genetic similarity with the recipient parent were selected. These lines exhibited high level of resistance against yellow rust under epiphytotic conditions. These superior lines which were foreground Yr marker positive with high levels of morphological similarity and recurrent parent genome similarity were selected for field trials. The study demonstrated successful application of MAS for targeted pyramiding of durable yellow rust resistance genes into the popular wheat varieties.
