Integration of Rice Zinc Finger Protein Gene into Eggplant (Solanum melongena L.) Genome

Abstract


The present study focused was on the optimization and stabilization of transformation protocols for eggplant (Solanum melongena L.) with the rice zinc finger gene. Tissue culture of pure verified eggplant variety seeds (NS797) was initiated with 25% coconut water content and eggplant shoot hypocotyls were used for Agrobacterium tumefaciens (EHA 105) mediated transformation process at an optimized concentration of 100µM of Acetosyringone, temperature of 26.5 °C. Hygromycine resistant gene harbouring vector (pCAMBIA 1301) was used for achieving maximum transformation rate of 15%. Optimum time for co-cultivation was established to be 24 hours. Standard OD 0.5 at 600nm was maintained for bacterial cultures that gave maximum transformation results. Stable transformations via Agrobacterium were observed in eggplant and PCR bands confirmed putatively stable integration of the zinc finger gene into the plasmid and the eggplant genome. Zinc finger gene for diseases resistance against variable range of insects was successfully integrated into the eggplant genome and that will prove to be a great push for sustainable growing of egg plants free from insect and pest attacks. Selection of transformants on selectable medium was confirmed by PCR.
