ELICITORS AFFECTS THE PRODUCTION OF PHENOLICS ON SHOOT CULTURES OF Hypericum amblysepalum Hochst

Abstract


In this study, we examined the effects of elicitors MJA and JA (50, 100 and 200 µM) on production of 15 phenolic compounds in the shoot culture of Hypericum amblysepalum Hochst. Phenolic compound determination was performed by LC-MS/MS method. The highest biomass production was obtained from shoots elicited with 50 µM JA (0.50 g L-1 DW), however increased JA concentration caused to a significant biomass inhibition. The application of elicitor MJA has a negative effect on the growth of the shoot cultures while led to an increase in the secondary metabolite content. The JA 200 µM treatment were increased the amount of the luteolin-7-glucoside (6.9 fold), apigetrin (4.1 fold), apigenin (2.9 fold), luteolin (2 fold), quercitrin (1.9 fold), and hyperoside (1.3 fold) compounds. The elicitor MJA(especially 200 µM) increased the secondary metabolite rutin (2.8 fold), astragalin (2.4 fold), protocatechuic acid (2.3 fold), luteolin (2 fold), hesperidin (2 fold), pseudohypericin (1.9 fold), apigetrin (1.9 fold), quercitrin (1.7 fold), chlorogenic acid (1.4 fold), hypericin (1.2 fold) and hyperoside (1.1 fold). With the recent study it is revealed that the shoot culture of H. ambysepalum through treatments two elisitors, it promises a very good source of raw materials for pharmaceutical industries.
