Thin layer chromatography of ergot alkaloids synthesized by Penicillium expansum

Abstract


Abstract The present study deals with the production of ergot alkaloids from Penicillium expansum using surface culture technique and estimation of ergot alkaloids was done using qualitative and quantitative techniques such as thin layer chromatography (TLC) and spectrophotometry. Different fermentation media were prepared, and pH was maintained at 5 with the help of 0.1N NaOH. These flasks were on shaker on 200 rpm at 25C temperature for five days. After that the optical density (OD) was measured at 590nm using spectrophotometer for the determination of ergot alkaloids. Best ergot alkaloids yield was obtained from the sample having yeast extract (2.73±0.01), (2.72±0.02) as compared to the sample having beef extract (2.708±0.02, 2.703±0.03) in the fermentation medium. During TLC analysis, various mobile phases were screened in the first phase such as A, B, C, D, E, F, H. In this phase, TLC plates revealed Van Urk reagent positive spots in mobile phases H (Chloroform 120: propanediol 30: water 20) that also gave a maximum Rf valuefor ergot alkaloids. In the next phase, mobile phase H, was further modified to obtain the maximum separation of ergot alkaloid contents present in the sample. Maximum Rf value was obtained from filtrates PEM3 and PES3 with Rf value 0.96, 0.95 in the mobile phase H2.
