IN VITRO PROPAGATION OF SUPERIOR MONOEMBRYONIC MANGO (MANGIFERA INDICA L.) VARIETIES OF PAKISTAN

Abstract


A successful somatic embryogenesis (SE) and plantlet regeneration method exploiting the nucellus tissue was established from three superior monoembryonic mango vars. Saroli, Langra and Chaunsa grown in Khairpur region, Sindh, Pakistan. Immature fruitlets (2.5-4 cm long) of three varieties were brought in the laboratory for surface sterilization using 30% NaOCl solution. Direct somatic embryogenesis (93%) as maximum was achieved successfully in var. Chaunsa under complete dark conditions on the medium comprising 2.0 mg L-1 2iP + 0.5 mg L-1 2,4-D. On the contrary, the medium comprising 4.0 mg L-1 2iP + 1.0 mg L-1 2,4-D induced the embryogenic callus (91%) in var. Chaunsa. The medium comprising micro salts of MS + macro salts of Gamborg’s B5 medium in addition to 0.1 mg L-1 2iP + 0.5 mg L-1 Kin revealed the significantly highest germination rate (95%) of somatic embryos in var. Chaunsa. Var. Langra showed the significantly highest response of increase in shoot length (5.1 cm) and root length (4 cm) on the medium comprising micro salts of MS + macro salts of Gamborg’s B5 in addition to 0.1 mg L-1 NAA + 0.2 mg L-1 BA. The varietal independent responses were observed among the studied varieties regarding the induction of somatic embryogenesis (direct and indirect), rooting and plantlet regeneration which may support to apply the developed protocol for in vitro propagation of other monoembryonic varieties of mango around the world.
