MOLECULAR OF RAPD MARKERS AND PHYSIOLOGICAL CHARACTERISTIC IN TOMATO INDUCED BY CHEMICAL MUTAGEN

Abstract


One effort to improve the quality of tomato plants is done through mutations. The local tomato was treated with EMS in three different concentrations (0, 0.3% and 0.5%) and three doses of potassium fertilizer (0, 2,5 g and 5 g) per plant. DNA isolation Kit was used for isolation DNA whole-genome of young tomato plant leaf. OPA primers 07, 08, 11, and 20 were used for DNA amplification. The results of the analysis using RAPD showed that genetic variation occurred. The provision of EMS at different doses resulted in genetic changes in the tentative yield of tomato plants, namely at EMS 0.3% and 0.5 % without addition of potassium. The effect of fertilizer and EMS was only significant on Reducing Sugar content, Vitamin C, Total Protein and fruit brix content. Using potassium at different doses has no effect on genetic changes in tomato plants. Expression of those primers formed 2 clusters in 4 DNA samples amplified from 4 primers. The primers used resulted in a polymorphism percentage of 62.3% from a total of 12 DNA bands. Induction of mutations with concentrations has not yet been identified.
