complete chloroplast genomes of two spatholobus species: molecular structures, comparative analysis and phylogenetic relationship in leguminosae

Abstract


The vine stem of Spatholobus suberectus Dunn (S. suberectus), called “JiXueTeng”, has used as a significant medicine for thousands of years in China. However, reliable field identification of this medicinal plant remains problematic, which may cause serious adverse effects in the functions of the drug and may affect the clinical medication reviews. To ensure the use exactly of medicine and implement protective legislation, it is imperative to obtain the chloroplast (cp) genome of S. suberectus, which can be used as the valuable resources for species identification and phylogenetic analysis. In this study, the complete cp genomes of S. suberectus (152,173bp) and Spatholobus pulcher (S. pulcher) (151,099bp) were assembled for the first time to gain the abundant information for the genus of Spatholobus by using the next-generation sequencing (NGS) technology. We found the GC content of S. suberectus and S. pulcher were closely, 35.19% and 35.37%, respectively. The noncoding region was more divergent than coding ones. Moreover, we revealed eight divergence hotspots (trnH, trnK-rbcL, trnL-rbcT, psbD-trnT, trnC-rpoB, atpI-atpH, ycf4 and trnL-rpl32) which might be used as candidate molecular markers for Spatholobus identification. The analysis of phylogenetic relationship indicated that two Spatholobus species were clustered together and was sister to Cajanus. The findings of this study were conducive to species identification and phylogenetic research of Spatholobus and provided valuable resources for exploration research on substitution of S. suberectus.
