LABORATORY CULTURE OF ULVA INTESTINALIS UNDER DIFFERENT SALINITY, TEMPERATURE AND ILLUMINANCE

Abstract


The reproductive pattern of green seaweed Ulva intestinalis was studied from 14th November to 28th November 2014 , and the reproduction of U. intestinalis were observed under the conditions with different salinities 20, 25, 30 ppt, 20, 40 µmol m-2 s-1 light condition, ,and different water temperatures 10 and 20oC. Asexual and vegetative phase of reproduction observed during the fourteen days of culture supplemented with 1mol/L NaNO3 and 0.1 mol/L KH2PO4 seawater. No sexual reproduction was observed, but gametes observed in 10 oC, 20 µmol m-2 s-1 and 20 ppt salinity respectively. The maximum SGR shown in (40 µmol m-2 s-1, 25 ‰ and 20 oC) 27.56% SGR with increased length of filament 11.93±6.64 mm. Proliferation of U. intestinalis thallus could vegetatively grow. The maximum size observed is 55.89±34.49 µm when cultured in 25 ppt, 20 µmol m-2 s-1 and 10oC respectively. During the asexual reproduction, the motile male and female zoospores was observed in 20-40 µmol m-2 s-1,10-20 oC, 20 and 25 ppt cultured medium. It is indicated that U. intestinalis grow fastly by vegetatively and asexually in all trial of culture experiment.
