identification of glutathione peroxidase like enzymes in wild watermelon (Citrullus Lanatus) and its expression analysis under abiotic stress

Abstract


Among abiotic stresses, drought is the major stress that effect plants which in turn affects their working capability and productivity. The increased level of drought results in the production of high level of reactive oxygen species (ROS). Antioxidants are agents that scavenge the adverse effects of oxygen produced in the form of ROS. Under water deficit conditions in Citrullus lanatus, level of Glutathione peroxidase-like enzymes (GPXL’s) raises abruptly which hydrolyses H2O2 using Thioredoxin (Trx). C. lanatus is xerophyte and native of Kalahari Desert, Africa. In this study GPXLs in C.lanatus were analysed to exploit their role in water use efficiency. To validate the presence of GPXLs, a conserved motif (signature 3) of GPXLs was amplified using PCR in wild watermelon. Furthermore, a novel GPXL gene, GPXL(w), was identified in C. lanatus using gene specific forward and reverse primers from cultivar species. The resulted sequence was blasted in NCBI which showed resemblence with GPXLs in other plant species e.g. Cucumis sativus probable glutathione peroxidase 5 (98.39%), Momordica charantia probable glutathione peroxidase 5 (93.5%), Ziziphus jujuba probable glutathione peroxidase 4 (83%), Citrullus lanatus cultivar Charleston Gray (80%). The localization of GPXL(w) gene was predicted in plastids using Bioinformatic algorithm. The phylogentic studies of GPXL of wild watermelon showed close relationship with GPXL4 of C. lanatus cultivar having 95% confident branch value. Additionaly, the GPXL of C.lanatus(w) is more closely grouped with GPXL5 of cucumber having branch value of 57%. The presence of GPXL(w) in wild watermelon suggests a key role in drought tolerance, however, identification of other isoforms of GPXLs is prerequisite for vital understanding of this gene family in wild watermelon.
