Callus Culture Extracts of Traditional Herb Withania somnifera Dun. Attenuate STZ-induced Diabetes in Rabbits

Abstract


Diabetes mellitus affects badly human of all ages in population and results in millions of deaths every year. This study was initiated to investigate the antidiabetic potential of callus cultures of Withania somnifera in Streptozotocin (STZ) induced rabbits. Hypocotyl and cotyledon explants from in vitro seedlings produced good callus on MS medium under plant growth regulator (PGRs) combination of 0.05 TDZ with 0.5 BAP mg/l. Among crude extracts chloroform extract of hypocotyl callus (HC) presented maximum percentage yield. Phytochemical analyses of the extracts indicated the presence of alkaloids, phenols and flavonoids with good visual rating. Chloroform HC extract in oral application at 100 mg/kg body weight of rabbits showed sustainability in body weight and antidiabetic effect comparable to standard dose of Metformin in STZ-induced diabetic rabbits. In brief, callus cultures appeared to hold promising potential to manage diabetes and offer an in vitro renewable source of biomass for extraction of phytochemicals and plant conservation as well.
