Effects of Aluminum on the Cell Morphology of Root Apices in Two Pineapples of Different Al-Resistance Characteristics

Abstract


Pineapple (Anonas comosus (L) Merrill) is a kind of economically cultivated crop which is commonly planted in strongly acid soil in Taiwan. However, aluminum (Al) in strongly acid soil is maybe toxic and inhibits the roots growth of some crops. This study was conducted to evaluate the effects of Al concentration (0 vs 300 μM AlCl3) on the root apices cells of Al-resistant Cayenne and non-resistant Tainung No.17. After treating with 300 μM AlCl3, the root apices cells of Cayenne was not affected while that of Tainung No.17 showed disintegration of inter-cellular layers when observed with an SEM (scanning electronic microscopy). Further observation under a TEM (transmission electronic microscopy) indicates no significant variation in the cells of root apices of Cayenne after treating with 300 μM AlCl3, however, apparent changes in the cells of root apices of Tainung No.17 were observed, e.g. the separation between the cell wall and cell membrane, the enlargement of vacuole, the shrink and the gathering of mitochondria beside inner cell membrane. The contents of callose (1,3-β-glucan) and MDA (malondialdehyde) in Cayenne were not affected by treatment of Al concentration, however, those in Tainung No.17 showed significant increases under the treatment of 300 μM AlCl3, indicating indeed damage on the root apices of Tainung No.17. The results served an important clue that the pineapple possessed different aluminum resistance should be planted in different soil environment.
