Current Status of Fusarium Head Blight in Northwest Pakistan: Survey for FHB Resistance using Molecular Markers.

Abstract
In spring of 2015-16, Fusarium Head Blight (FHB) was observed in fields of Swat District in Northwest Pakistan. The Initial surveys based on symptoms revealed that the incidence ranging from 34-84%, is limited to few fields, growing wheat cv. Ata Habib, along River Swat in Tehsil Kabal. Keeping in view the environmental factors in Northwest Pakistan around spring, the disease has great potential to reach epidemic proportion, if left unchecked and may lead to devastating losses both in terms of quality and quantity. As a prophylactic strategy, we carried out a survey of elite Pakistan wheat cultivars and CIMMYT lines for FHB resistance using molecular markers specific for FHB-1, 2 and 3. Among the Pakistani varieties, Marvi-2000 showed presence of FHB-2 and 3 specific bands and Saleem-2013 amplified FHB-1 and 2 specific bands, respectively. While among the CIMMYT lines only line 111 amplified bands specific to FHB 1 and 3. Almost all the CIMMYT lines show FHB-1 resistance. Furthermore, all the germplasm was screened for 2NS Translocation - from Triticum ventricosum segment contain cluster of resistance genes for many diseases including FHB. 2NS translocation was validated in 2NS near isogenic lines (NILs) obtained from Kansas State University using 2-NS specific marker VENTRIUP and LN2 , however, no 2-NS translocation was found in Pakistani varieties and CIMMYT lines. Among 2NS-NILs, Yaccora-Rojo-2NS (YC-2NS) not only amplified 2NS specific band but also FHB1, -2 and -3 specific bands. Our findings will provide a baseline for further research on FHB and breeding strategies to introgress the available resistance into elite cultivars.
