ANALYSIS OF DIVERSITY AND RELATIONSHIPS AMONG IRANIAN NATIVE SALICORNIA VARIETIES USING AFLP MOLECULAR MARKER

Abstract


Premise: Salicornia is a halophyte that has been introduced in many countries as a strategic plant due to its unique characteristics such as high resistance to salinity and using saline water for irrigation.Scrutiny of the species according to morphological characteristics alone is not simple; This study is performed to assess the genetic diversity of Iranian Salicornia species compared to European Salicornia species using AFLP markers. Method: In this study, Salicornia species were collected from different parts of Iran. The genomic DNA of 21 different varieties belonging to four different Salicornia species and Halocnemum control species were studied by using AFLP marker and by a combination of EcoRI / MseI. the cluster grouping classified the species based on UPGMA method by using Jaccard similarity coefficient. Results:4 Salicornia species studied completely separated from each other at a genetic distance 31% and were divided into 5 groups. In this dendrogram, the samples were isolated with sufficient evidences and arguments. Conclusions: there is a relationship between genetic diversity and geographic distribution and AFLP markers confirmed the results of morphological studies on similar species in Iran. So the central regions of Iran can be considered with high confidence as one of the habitant of Salicornia.
