IN VITRO SCREENING AND EVALUATION OF VARIOUS CALLUS CHARACTERISTICS BY PHYSICAL STATE TO IMPROVE REGENERATION EFFICIENCY IN POTATO (SOLANUM TUBEROSUM L.)

Abstract


Potato (Solanum tuberosum L.) is one of the most important food crops capable of nourishing the world’s population. To improve potato through biotechnological approaches a suitable callus inducing genotypes and an efficient regeneration systems, ten indigenous and six exotic genotypes were taken for this study. Internodal segments of those genotypes were cultured on MS medium supplemented with 3.0 mg/L 2,4-D + 0.5 mg/L Kin. Various physiological parameters such as percentage of callus induction (PCI), relative fresh weight growth (RFWG), relative growth rate (RGR), callus growth rate (CGR), callus fresh weight (CFW), callus dry weight (CDW) and relative water content (RWC) were considered for the evaluation procedure. Data were recorded on the basis of all indices mean rank and standard deviation of ranks. ANOVA, DMRT and correlation co-efficient on all in vitro callus inductions characteristics were found highly significant (p <0.01) among the tested genotypes. In this case, Surjamukhi, Granola, Sheelbilati, Arun and Sindurkouta exhibited the good callusing (ranges of rank sum were 5.07 to 10.17) rate while Courage, Diamant, Jamalu, Sadaguti, Patnai and Cardinal showed fair callus inducing genotypes (10.88 to 13.96). On the other hand Lalpakri, Asterix, Chollisha, Dohazari and Ausha performed poor callusing. When calli of these genotypes were cultured in MS + 5.0 mg/L BAP + 2.0 mg/L IAA + 1.0 mg/L GA3, highest shoot regeneration frequency occurred in Sadaguti (40%).
