SIMPLE SILVER STAINING PROCEDURE FOR DNA MICROSATELLITE POLYACRYLAMIDE GEL ELECTROPHORESIS

Abstract
Because of its complicated, multi-step procedure and large time requirements, silver staining procedure of PAGE has become a rate-limiting step in the detection of DNA fragments in polyacrylamide gel. Many alternatives to silver staining methods have been described, but these methods are not efficient or cost-effective. A simple and efficient method of silver staining was established in this study. The procedure comprises only four simple steps: rinsing, staining, rinsing, and development. It can be completed within 15 min. The method does not require the preparation of fixer solution and developer stop solution. In addition, two rinsing steps only using tap water can achieve satisfactory results.
