PHENOTYPIC AND MOLECULAR ANALYSIS OF SEGREGATING POPULATIONS OF WHEAT FOR STRIPE RUST (YELLOW RUST) RESISTANCE

Abstract
The rsearch was initiated by developing five populations (P1,P2,F1,F2 and F3) of the seven cross combinations of wheat including RK-23 × Maxipak, RK-23 × Inqilab, RK-23 × Seher, RK-23 × Zincol, RK-23 × Shahkar, RK-23 × Borlague and RK-23 × Gold. The Five populations of each cross were sown at two locations i.e Agricultural Research Institute, D.I.Khan and CCRI,PirSabak, Nowshehr. Data were collected on rust resistance traits including reponse value, severity percentage and coefficient of infection of yellow rust. Molecular study was also conducted by using three molecular markers for the screening of genotypes in F3 generations for the presence of rust resistant genes i.e Yr15, Yr17 and Yr18 genes. ANOVA results shows the significant difference between both the environmenatl condition for all the yellow rust traits . The generations of all the cross combinations showed highly significant difference for the rust traits except RK-23 × Seher and RK-23 × Zincol. Significant value of generations × location interaction was found in all cross combinations except RK-23 × Zincol for all the rust disease traits. The highest mean for reposnse value of yellow rust (0.93), yellow rust severity(56.8%) and coefficient of yellow rust (46.5) was noted in Inqilab variety and its sucessive generations in cross combination RK-23 × Inqilab i.e F1-F3. The lowest mean for response value (0), severity percentage (0%) and coefficient of infection of yellow rust (0) was recorded in Shahkar variety and its sucessive generations in cross combination RK-23 × Shahkar. Molecular analysis study shows that Yr17/Lr37/Sr38, Lr34/Yr18 and Yr15 genes were detected in some of the F3 lines of cross combinations RK-23 × Zincol, RK-23 × Shahkar and RK-23 × Borlague. Therefore, on the basis of performance for phenotypic and molecular data regarding rust resistance, the populations of cross combinations RK-23 × Shahkar, RK-23 × Zincol and RK-23 × Borlague should be continued in rust resistance breeding.
