IDENTIFICATION AND VALIDATION OF RELIABLE REFERENCE GENES IN SAPOSHNIKOVIA DIVARICATA UNDER LOW-TEMPERATURE STRESS

Abstract
In this study, we selected and validated reference genes from Saposhnikovia divaricata under low-temperature stress based on transcriptomic data. This research employs qRT-PCR experiments and computer software tools, such as geNorm, NormFinder, BestKeeper, Delta Ct, and RefFinder, to recognize stably expressed reference genes in leaves, stems, and roots. The reliability of these preferred genes was confirmed based on analyzing variations in the expression of the sucrose phosphate synthase (SPS) gene under low temperatures. Therefore, these results are of great significance for prospective studies that aim to analyze changes in the content of active substances in S. divaricata under abiotic stress, enhancing qRT-PCR normalization and promoting further research on gene expression.
