Genetic diversity analysis of peony germplasm resources based on ISSR markers

Abstract: To uncover the genetic correlation among peony germplasm and enhance the efficient safeguarding and exploitation of peony germplasm resources. In this study, ISSR molecular markers were used to analyze the genetic diversity and genetic relationship of 45 peony samples collected from Yaowangshan Peony Garden in Luoyang, and UPGMA method was used for cluster analysis. The results indicate that 12 ISSR primers were employed to amplify the genomic DNA of 45 peony varieties, resulting in a total of 106 polymorphic loci. The average number of bands detected per primer was 8.83, with an average percentage of polymorphic loci of 93.25%.Cluster analysis by UPGMA method showed that the genetic similarity coefficient of the tested materials was between 0.5566 and 0.9151, and the tested materials were divided into 7 clusters, indicating that ISSR markers had rich polymorphism among peony varieties and could well reveal the genetic relationship among peony varieties. The results of this study can provide theoretical basis for variety identification and hybrid breeding of peony germplasm resources.
