A cytological study on the meiosis process of Humulus scandens

Abstract
Humulus scandens (HS) is a common dioecious weed that possesses valuable forage and medicinal properties. It exhibits strong adaptability and has a high seed yield, making it a subject of significant research interest. However, there is a lack of information regarding the meiosis process of HS and the relevant methods used for studying it. In this study, we aimed to address this gap by utilizing intact HS meiosis cells as research material. We employed a rapid labeling method to observe the meiotic chromosomes. Furthermore, a single oligonucleotide probe of 5S rDNA was used to determine that the number of 5S rDNA signals in the somatic cells of HS was one pair. Based on this probe, we conducted an inspection of homologous chromosome pairing in HS. Our study provides a comprehensive description of the meiosis process in HS and presents a set of cytological methods that are suitable for easy labeling of meiotic chromosomes and monitoring homologous chromosome pairing in HS.
