COMPARISON OF BIOCHEMICAL COMPONENTS AMONG THE MUTANT AND PARENT RICE CULTIVARS- A COMPARATIVE CROSS-SECTIONAL STUDY

Abstract


Background: Rice is the second most widely consumed cereal in the world next to wheat, while it is the main cereal in celiac patients. In recent years, rice cultivars have demonstrated revitalization in various agronomic characteristics, so it is also essential to consider the nutritional aspects. There is a dire need to evaluate the nutrient content of different rice varieties so that appropriate can be taken to fulfill the nutrient requirements of the consumers. Therefore, this study was conducted to compare the nutrient content of different rice varieties available in Pakistan. Materials and Methods: Thirteen rice cultivars were collected from the Nuclear Institute of Agriculture (NIA) Tandojam, Sindh, Pakistan.  The rice varieties were evaluated for determination of total moisture, total ash, amylose, and total protein content including different protein fractions, albumin, globulin, prolamin, and glutelin extracted in different buffers by the standard method. The SDS-PAGE gel electrophoresis was performed for the screening of protein fractions. Data were analyzed by using Graph Pad Prism version 7.0 and Microsoft Excel 2016. Results:  The study reveals that rice varieties under study showed a significant difference among the parent and mutant varieties. The moisture content and ash content were higher in parent varieties as compared to mutant cultivars, while protein content was higher in mutant varieties (p-value <0.05). Conclusion:  The nutrient content was in general good in all varieties, though parent varieties showed better contents. The nutrient content is influenced by the environment, storage, and how rice is consumed. Further large-scale studies are recommended to compare rice varieties considering these factors.  Key Words: Rice, screening of rice fractions, moisture, ash, protein, and amylose.
