UV-IRRITATION MEDIATED DEVELOPMENT OFASPERGILLUS NIGER GCB-117 STRAIN FOR HYPERP RODUCTION OF CITRIC ACID

Abstract
One of the most significant industrial metabolic products produced by microbes is citric acid. Aspergillus niger is regarded as one of the best citric acid producers, among other sourcesFor enhanced production, it is important to develop strains with superior metabolism. In this study, we have exposed Aspergillusnigerto UVradiations to obtain a high yield of citric acid from UV-induced mutant strains.Atotal of 54 mutants were screened for higher production of citric acidin a shake flask containing potassiumferrocyanide-treated sugarcane molasses. Mutant strain GCB-117UV-52 produced 17.0% more citric acid (23.4 g/l) as compared to wild strain GCB-117 (20 g/l). This mutant was further screened for enhanced citric acid production against various cultural conditions. Maximum yield (24.5 g/l) was obtained when potassium ferrocyanide (200 ppm) was added at 0 h after inoculation. Citric acid production was induced (35.3 g/l) by adding 3.0% methanol to the fermentation medium. NH4NO3 (0.20%) was optimized for maximum yield (42.6 g/l), which was found to be 82.05% increased level as compared with that obtained before optimization.
