Soil microbiota and Identification of microorganisms using 16S rRNA gene sequencing by Illumina Miseq in the Hail region of Saudi Arabia

Abstract


This study mainly focused on the possible bacterium types that could be found in soil samples extracted from the Hail region (‏27°59’05.7” N and 41°45'46.3” E), in north-central Saudi Arabia. Microbiota (microbial population) have influence on human, and animal immunity, geochemical nutrient cycles, and plant health and productivity. The association of microbes with soil and with the native plant in its habitat play a prominent role in the development and identification of various diseases and their pathogenesis. Metagenomics has evolved a unique technique for the isolation of DNA from environmental samples such as soil without culturing these microbes. In this study, we used the 16S rRNA gene-based metagenomics analysis by Illumina Miseq to investigate the impact of microbial communities found in farm soil using NCBI nucleotide collection. Highly similar sequences were subjected to Megablast program and retrieved sequences were matched with the query sequences. Examination of more than 1000 sequences revealed >95% significant alignment was found in 600 sequences and 100 taxa had pathogenic (35 species), non-pathogenic (18 species), parasitic (18 species), and while, 4 unidentified bacterial species were also found. The results of the study reveal that a diverse population of bacterial strains was found and soil microbiome dynamics might be a soil indicator, as well as pathogens related to plants, and helpful for the identification of favourable microbes for plant growth. Thereby, the main aim of this study was to identify the bacterial species that are pathogenic to plants using novel molecular techniques. Further more research is needed to investigate the actual relationship between the isolated bacterial species and their ecological habits as well as to identify their medicinal value.
