Natural Neuroprotection: Investigating the Effects of Solanum nigrum Extract on ATP-Induced Cell Proliferation in PC12 Cells

Abstract


The importance of natural products in drug discovery has grown significantly, leading to increased efforts to screen for effective drugs from natural sources. S. nigrum, also known as black nightshade, has been reported to possess anti-proliferative, antioxidant, antimicrobial, and anti-inflammatory effects. In this study, we investigated the effects of S. nigrum extract on ATP-induced cell proliferation in the PC12 cell line. Our results showed that treatment with S. nigrum (30 µM) led to a decrease in cell proliferation and an increase in cell death, as measured by cell counting, LDH assay, and H33258 staining. However, S. nigrum (30 µM) was also found to significantly ameliorate the toxic effects of ATP (100 μM) on the PC12 cell line. These findings suggest that S. nigrum extract has a neuroprotective effect on the PC12 cell line after 72 hours of treatment. While the detailed effect of S. nigrum on the calcium-mediated pathway is still under analysis, our preliminary data suggest that it could be an effective neuroprotective bio compound.
